Speaker Biography
Dr. Rob Klose did his PhD with Professor Sir Adrian Bird at the University of Edinburgh where he studied proteins that bind to methylated DNA. He did his post-doctoral training with Professor Yi Zhang at UNC Chapel Hill where he identified histone demethylase enzymes and studied their roles in gene regulation. Rob is now the Professor and Chair of Genetics at the University of Oxford, where his group is focussed on understanding how CpG island elements function to regulate gene expression.
Title
Dissecting Polycomb-mediated gene repression
Abstract
Polycomb Repressive Complex 1 (PRC1) and PRC2 function through chromatin-based mechanisms to repress gene expression and are essential for normal mammalian development. However, how PRC-dependent repression is achieved at the molecular level has remained poorly defined. Addressing this problem has been complicated by the interrelatedness of PRC1 and PRC2 activities at target gene promoters. Here, I will discuss our efforts using rapid perturbation coupled with time-resolved quantitative genomic approaches to examine the central determinants of Polycomb-mediated gene repression in mouse embryonic stem cells (ESCs).
